Since MacPorts is configured to build for the x86_64 architecture where possible and psyco can only be built for i386, you will probably need to run python like this to use psyco:
arch -i386 /opt/local/Library/Frameworks/Python.framework/Versions/2.6/Resources/Python.app/Contents/MacOS/Python
